I got the Kindle as a gift and compared it to other e-readers on the market. I'd pick it over the Nook, but it depends on what you want.
There are other e-readers besides these, too - and depending on what you want it for, might even be a better fit yet.

I have heard great things about the Kindle. They look great. I would buy one of them.

I have kindle. Love it. Better tech and make, hands down.

I got a Kindle touch for Christmas and its working great so far. The only complain I have is that sometimes, when I try to go back a page, it will skip to a random page.

Nook is generally overpriced and if you were going that far you might as well invest in a tablet.

I have a Nook because a family member works for the company which puts it out, and I got it as a gift. It's a hell of a lot better than a KOBO (the ill fated eReader Borders put out right before they declared bankruptcy) You can't get KOBOs fixed to save your life and they don't work properly (my screen just died and the WiFi on it stopped working after only a month and I was told after days of trying to get hold of KOBO that they had discontinued the WiFi feature and I had to through Adobe to get the books, after about 1,000 steps. I never bought an other book from them, and then my screen died.
I was happy to get the Nook. Although, I don't use it a whole lot. I do get some library books on it, but our system doesn't catalog them properly, so you have to go through alphabetical order to find a book. *sigh*
I've never even seen nor used a Kindle in person, so I can't really vote. I just know the NOOK is better than the KOBO.

This year for Christmas, my sister got a Kindle Fire from her in-laws and I got a Nook Tablet from my bf. Side by side, I prefer the Nook. While the Fire has 8G of usable storage, the Nook has the ability to use a micro SD card for additional storage space (it only comes with 2G I think).
But, I think it really depends on what you have or have have had in the past as to which you prefer. My sister really likes the Kindle Fire, even though she isn't able to fully use it since they do not have WiFi in their home.

NOOK COLOR...color still the way to go if you enjoy magazines, maps anything other then just books. For plain old books...with ZERO illustrations, go with Kindle. Easier on the eyes.

I have a Kindle Touch. I would advise against the particular model. The touch screen is super sensitive. Sometimes, when I am trying to turn to the next page, I will wind up a chapter ahead instead.

I'm planning on getting a Nook, but that's just because I have all these Barnes & Noble gift cards lying around and I may as well put them to good use. If that wasn't a factor, I'd get a Kindle hands-down.

Kindle I think the e-ink is very good and the battery last weeks

I have a Kindle Fire, and I love it... for the most part. My only complaint is that the "user guide" and help directions are useless. As for storage space, while there isn't an SD slot, you can get Cloud space from Amazon.
